Human mycoses: drugs and targets for emerging pathogens.

چکیده
Fungal infections in humans range flom superficial and cutaneous (such as dermatophytoses) to deeply invasive and dissemi hated (such as candidiasis and cryptococcosis). Pathogenic fungi occur worldwide, although particular species may predominate in certain geographic areas. In the past 20 years, fungal infections have increased dramatically--paradoxically, as a resuh of medical advances. Fungal infections occur more frequently m people whose immune system is suppressed (because of organ transplantation, cancer chemotherapy, or the human immunodeficiency vires), who have been treatM e m b r ed with broad-spectrum Polyen~

Exploiting Fungal Virulence-Regulating Transcription Factors As Novel Antifungal Drug Targets
Systemic and invasive mycoses caused by primary and opportunistic fungal pathogens have been emerging as global problems because of the increase in the number of immunocompromised individuals, due to solid-organ transplants, anti-cancer chemotherapy, and extended human lifespan. A recent report estimated that fungal pathogens, such as Cryptococcus neoformans, Candida albicans, and Aspergillus f...
متن کاملA Case for Two-Component Signaling Systems As Antifungal Drug Targets
The recent outbreak of fungal meningitis caused by Exserohilum rostratum in patients receiving contaminated steroid injections resulted in 64 deaths, receiving a lot of press and briefly bringing into the public eye the difficulty of treating systemic fungal infections [1]. What is generally less well appreciated, however, is that there are several other, much more common fungal pathogens that ...
متن کاملCandida Infections, Causes, Targets, and Resistance Mechanisms: Traditional and Alternative Antifungal Agents
The genus Candida includes about 200 different species, but only a few species are human opportunistic pathogens and cause infections when the host becomes debilitated or immunocompromised. Candida infections can be superficial or invasive. Superficial infections often affect the skin or mucous membranes and can be treated successfully with topical antifungal drugs. However, invasive fungal inf...
متن کاملEmerging disease issues and fungal pathogens associated with HIV infection.
Fungal diseases are increasing among patients infected with human immunodeficiency virus (HIV) type 1. Infections due to Candida and Cryptococcus are the most common. Although mucocutaneous candidiasis can be treated with oral antifungal agents, increasing evidence suggests that prolonged use of these drugs results in both clinical and microbiologic resistance. The optimal therapy for cryptococ...
متن کاملFungal Glucosylceramides: From Structural Components to Biologically Active Targets of New Antimicrobials
The first work reporting synthesis of glucosylceramide (cerebrin, GlcCer) by yeasts was published in 1930. During approximately 70 years members of this class of glycosphingolipids (GSL) were considered merely structural components of plasma membrane in fungi. However, in the last decade GlcCer was reported to be involved with fungal growth, differentiation, virulence, immunogenicity, and lipid...
